Production of AAV using the AAV-MAX system in a 3 L stirred-tank bioreactor

Adeno-associated viruses (AAVs) are increasingly applied viral vectors in gene therapy due to their safety, low immunogenicity and long-term transient expression. As a result, it is necessary to effectively scale-up AAV vector production methods to satisfy this growing market. It is crucial for such processes to deliver high titers in a scalable and cost-effective manner. The Gibco™ AAV-MAX Helper-Free AAV Production System can be used to produce AAVs in the Thermo Fisher Scientific™ HyPerforma™ Glass Bioreactor. Optimizing bioreactor process parameters provides growth kinetics and metabolite profiles, as well as reproducible AAV2 and AAV6 titers, that are equivalent or superior to those achieved in shake flask production.
